762 Analytical method to monitor squalene and its primary oxidation products by liquid chromatography coupled with both mass spectrometry and diode array detector

Squalene (SQ) is a lipid produced in sebaceous gland involved the vital skin barrier function. Its peroxidation leads to accumulation of SQ epoxides and monohydroperoxides causing various damages as hyperpigmentation, wrinkles, acne. Monitoring effects cosmetic cream on evolution content their oxidation during clinical studies represent numerous analysis; highlighting need combine molecule analyses single method. High Performance Liquid Chromatography (HPLC) coupled with Diode Array Detector (DAD) commonly allowed detect but not sensitive enough for detection products. On other hand, structure cannot be detected by Mass Spectrophotometer (MS) although it more sensitive. The aim this study was propose method analyze its products within injection coupling DAD MS analyses. First, standard oxidized hydrogen peroxide. Separation unoxidized developed HPLC-DAD-MS. It at 210 nm identification electrospray ionization negative scan mode m/z=409 m/z=425.
